Ex vivo precision-cut organ slices offer an unparalleled model to evaluate human tissue in numerous research settings…therapeutics, toxicology, physiology, disease progression, etc. While tissue slice work has been conducted for decades, recent advances in culture methods, preservation technology, and detection of complex endpoints has positioned organ slices as a highly physiological, and human relevant test system. The inclusion of all cells types in the tissue (including immune cells), native architecture, and ability to culture for weeks or months makes this model highly desirable for new approach methodologies (NAMs).
Precision-cut Lung Slices
Precision-cut lung slices (PCLuS) have been regarded as perhaps the most physiologically relevant human-derived model of the deep lung. Containing all cell types (including alveolar macrophages, etc.) in the tissues at the time of slicing, PCLuS are highly immune responsive, present a native architecture (complete with alveolar and small airway structures), and can be maintained for 4 weeks or more. The long-term retention of relevant cell types and complexity, and widespread use in numerous applications give PCLuS an advantage over other in vitro tools used to study lung diseases and test the effects of various exogenous agents on respiratory health.
